what's up everyone it's niels again and
in this video i will show you the
protocol that i will be using to assess
protein digestion by trypsin there is
quite a bit of chemistry involved with
this protocol and as you will see
there's a number of steps that you will
have to pay attention to to ensure the
success of this experiment alright
enough talking let's get started
first this is what you will need to
perform this experiment a set of five
disposable culture tubes a sharpie to
write the id of each tube di water the
solution of one percent trypsin and a
solution of point zero one percent bapna
in addition to this you will need some
disposable pipettes a clamp to handle
the tubes when using the hot plate and
here i'm following the protocol
described in the human emp lab manual by
maryab and smith as usual you will also
need a lab coat some gloves and a pair
before doing anything i would highly
recommend turning on your water bath
this thing can take a little while to
get up to temperature and turning it on
right now will save you some time here
also you may want to turn on your hot
plate since we will be using it shortly
now we can label all the tubes so we can
identify them when we prepare the
different solutions
according to the protocol described in
the manual we want to label the tubes 1t
each tube represents a specific
treatment including 2 controls and 3 tests
tests
now that everything is labeled let's
proceed to the experiment in tube 1t you
want to add about 3 drops of di water
and 3 drops of trypsin and just for a
good measure don't forget to cap your
tube to prevent accidental contamination
moving on to tube 2t the second control
add 3 drops of di water and 3 drops of bapna
in tube 5t add three drops of trypsin
because we will incubate these tubing
ice we want the trypsin to be as cold as
possible before adding ba pna that's why
we partially prepare it in advance
before we get back to it once all the
other tubes are ready
now we can prepare tube 3t add 3 drops
of trips into it and then if water is
boiling on your hot plate grab the tube
with the clamp and drop it in the
boiling water let it sit for about 5 minutes
meanwhile we can prepare tube for tea
add three drops of trypsin and three
drops of vip and a and as usual don't
it's been five minutes now so we can
turn the hot plate off and we can take
tube 3t out of the boiling water to put
tube 3t is almost complete add 3 drops
remember tube5t now that it's nice and
cold we can take care of it add three
drops of ba pna to it and like the other
we can finally proceed to leaving all
the tubes except tube5t in the water
perfect it's been about an hour now so
we can take all our tubes out of the
water bath to look at the results

disposable culture tubes a sharpie to
write the idea of each tube di water a
solution of one percent pancreatin some
litmus cream which is basically whipping
cream to which litmus powder has been
added to turn the cream blue and some
bile salts
in addition to this you will need some
disposable pipettes a clamp to handle
the tubes when using the hot plate a
micro spatula and some ice here i'm
following the protocol described in the
human emp lab manual by mariam and smith
as usual you will also need a lab coat
some gloves and a pair of glasses
before doing anything i would highly
recommend turning on your water bath
this thing can take a little while to
get up to temperature and turning it on
right now will save you some time here
we want to set it to 37 degrees celsius
also you may want to turn on your hot
plate since we will be using it shortly
now we can label all the tubes so we can
identify them when we prepare the
according to the protocol described in
the manual we want to label the first 5
tubes 1l 2l and up to 5l
the last two tubes should be labeled 4b
and 5b
each tube represents a specific
treatment including two controls and

proceed to the experiment
in tube 1l you want to add about 5 drops
of di water and 5 drops of pancreatic
and just for good measure don't forget
to cap your vial to prevent accidental contamination
moving on to tube2l the second control
add five drops of di water and five
in both tubes 5l and 5b add 5 drops of
paint creating because we will incubate
these tubes in ice we want the paint
creatine to be as cold as possible
before adding litmus cream and vitals
that's why we partially prepare them in
advance before we get back to them once
now we can prepare tube 3l add five
drops of pancreatic to it and then if
water is boiling on your plate grab the
tube with the clamp and drop it in the
boiling water let it sit for about 5 minutes
meanwhile we can prepare 2 4l add 5
drops of litmus cream and 5 drops of
pancreate and as usual don't forget to
we can also prepare tube 4b by also
adding five drops of fitness cream and
five drops of pancreatic the only
difference with cube 4l is that we will
once added make sure to stir the
it's been five minutes now so we can
turn the hot plate off and we can take
tube 3l out of the boiling water to put
it back in the holder
tube 3l is almost complete add 5 drops

remember troops 5l and 5p
now that they are nice and cold we can
take care of them into 5l add three
drops of liquid cream and like the other
in tube 5b also add 5 drops of litmus
cream but on lightroom 5l we will also
add a pinch of bile salt to it
once added make sure to stir the
solution a bit to mix all the chemicals
we can finally proceed to leaving all
the tubes except to 5l and 5b in the
water bath for about 1 hour
all right it's been about an hour now so
we can take all of our tubes out of the
water bath to look at the results
